We want to measure protein expression levels of SirT1 in mouse tissue (heart, skeletal muscle, liver) via Western blot. Unfortunately, no antibody has worked so far. Can someone recommend a working antibody and tell his protocol?
Hopefully this may work. I have attached a reference paper, they have tried the antibody in mouse tissues, you may want to increase your protein loading to >70ug. Good luck.
I would recommend a free antibody database available at labome.com. For SirT1antibodies you can check the following link: https://www.labome.com/review/gene/human/SIRT1-antibody.html. There you can find antibodies from Santa Cruz, EMD Millipore and Cell Signaling Technology used in WB for knockout validation. In particular, Santa Cruz offers rabbit polyclonal (H-300) used for WB on murine samples (ref: Fu et al., Mol Cell Biol. 2014). Also, Abcam has ab12193 suitable for WB on mouse samples. EMD Millipore has 07-131 ab which was used in western blot on mouse samples at 1:500 (ref: Heyward et al., J Neurosci. 2016). In addition, Cell Signaling Technology provides 9475 ab for WB on mouse samples at 1:1000 (Wang et al., Sci Rep. 2016). Sigma-Aldrich also has HPA006295 ab for WB on mouse samples.
