Can anyone point me toward a simple assay for either succinate or succinate dehydrogenase? I'm looking for something that I can learn and then work with an undergraduate student within a limited time frame. In the beginning, the source will probably be bone marrow macrophage cultures or J774 cells. Appropriate positive controls would also be most appreciated. A 700 dollar Elisa kit is probably not the way I want to go, if possible.
Hello, stranger! I am also struggling with this question. The kits are the simplest option, but other methods include NMR or mass spectrometry (with or without carbon-13), RT-PCR for one of the SDH genes (granted, expression does not necessarily equal activity), or a modified version of the DIY colorimetric assay from this paper:
