I am studying the Nlrp3 in brain. I chose the BV2 cell lines, and fortunately, I have detected the IL-1beta mature form in the SN after the LPS/ATP(1ug/mL and 2mM, respectively) treatment. Besides, I used the MTT to detect the cell viablity. Although the LPS did not induce BV2 cells dead, ATP(2mM, 1h) induced cytotoxicity. So, should I decrease the conc. or the time of ATP treatment ?
