ALT is a biomarker of liver damage, generally used to investigate hepatoprotective activities. Usually this marker is quantified in plasma/serum, but sometimes, it can also been assessed in liver homogenates.
1- May you share with me some publications reporting ALT assay in liver homogenates, After a chemical injury ?
2- using apap as an example of hepatotoxicant, Do you think ALT values in liver should increase or decrease. Because I saw some conflicting publications on that.

I think an increase in the level of ALT because this enzyme intracellular and any hepatotoxicants can lead to breakdown hepatocytes membrane and then release ALT to blood.
ALT levels rise in the plasma due to necrotic cell death where the cytosolic ALT can then traverse to blood plasma. Therefore, measuring ALT in blood is a metric of cell death. Measuring ALT in ground up liver tissue will not provide information about cell death and is only useful if you are interested in measuring ALT levels in liver.
Ok but the increase in serum may not necessarily indicate damage of liver tissue because ALT is also found in other tissues.
After induced- paracetamol liver injuries, surely the levels of ALT increase.
i advise to you to assay livers antioxidative enzymes, such as SOD or superoxide dismotase, or malone dialdehyde.....
This is a good marker for studying the role of the liver in several diseases
