After obtaining the total extract from the plants, our team is about to begin the separation steps using TLC. At first we need to know which solvent/s is the best solvent/s for separation of all chemicals in the total extract. the plan is to use TLC papers first and finding the best solvent/s to see the movements and bonds and then going over TLC plates to isolate and get the peaks.
finding the right solvent is a little time consuming, Does anyone can help us in this part? especially for separation of Alkaloids... which solvent or solvent mixture is the best to see the separated bonds on TLC paper?

You have to choose the best solvent to get Rf more than 0.4 starting from non-polar solvent e.g. hexane or toluene then mixture with polar solvent e.g. ethyl acetate or ethanol using different ratios 9:1, 8:2, 7:3 and so no till you get on the best ratio
