i am working on screening of wheat entries against aphids under controlled conditions. could anyone help me to clarify me on that it is standard protocol to add susceptible and resistant check for screening of plants against any insect, but if i had only susceptible check could it be used as a standard to evaluate my lines. or its obligatory to add resistant check also

I would not call it "mandatory", but rather "highly recommended" the inclusion of resistance wheat to this analysis. However if not available you can still assess differences between intermediate resistant plants through phenotypical screening.

In my point of view, if you have both resistant and susceptible check then it would give the accurate result in the experiment. Then it will be easy to scale the level of resistance in the test entries.
