It is very hard to use RNAi to knockdown the non-coding RNA transcripts that are only present in the nucleus of human cells for the lack of functional Dicer and AGO2 in the nucleus. You could use Cas13a for RNA targeting and it might work well in your study.
SiRNA-based approaches seem to be not useful regarding the artificial reduction of nucleus-located RNAs. Therefore, a knockout of the genetic information seems more adequate.
