I'm going to be sequencing some Poly(A) selected RNA and I was wondering if people have a RIN value they like to shoot for? Obviously higher is better normally, but since all the ribosomal RNA should be depleted in these samples I don't think a 18S/28S ratio is particularly informative. The RIN algorithm is proprietary and the only info I could find regarding RIN calculation, aside from the rRNA ratio, is that the chromatogram is split into 9 regions and assessed from there. As such, does anyone have an experimental correlation between a RIN score and "good" sequencing data?
