I recently ran a multiplex assay examining GM-CSF, IL-6, IL-8, IL-10, IL-1β, and TNF-α. After analyzing the data, I saw different results for my IL-6 and IL-8 than when I previously ran these same samples with an In-House ELISA. The significant trend observed in the ELISA is no longer present in the multiplex. I am trying to figure out why this is happening. I have contacted the company, but I am trying to determine why the results differ.
I would like to note that in the ELISA, I used a 1:5 dilution (20 μL of sample), which was done with fresh samples.
The multiplex was run neat with 25 μL of sample, and the samples had undergone two freeze-thaws before being run.
use same dilution. and add equal microgram or micromolar amount of sample. If you r using serum, then add after you measure total protein concentration. If all this not worked, try to wash well properly which will not lead any background noise. Check all the buffer comes with kit, eithr it is contaminated or not.
