To my understanding, Zn2+ in combination zinc ionophore blocks the replication of RNA viruses through the inhibition of RNA polymerase activity.

A recent study in Plos One (Velthuis et al., 2020) clearly presents the mechanism of action of the Zn ions and Zn-pyrithione ionosphore combination on the SARS-COV replication machinery.

Meanwhile according to Vincent et al. (2005) chloroquine (a quinine molecule) interfers with the ACE pathway.

Therefore, if quinine combines with zinc and zinc ions to elicit the same effect as observed in the Zn2+ / Zn-pyrithione combination, then a replacement will not confer the same results as observed with chloroquine.

However, the two options confer both prophylactic and therapeutic effects.
