I've been trying hardly to activate pyrin inflammasome in BMDMs but all my attempts did not work.

I will be glad if someone can help me solve this issue by giving me some tips or clarifying any missed step in the protocols I've followed so far:

Day-5 BMDMs were used in my experiments. 

Cells were plated in complete DMEM media (not containing M-CSF) for 30 min or 24h before priming.

Priming was then done with LPS (20 ng/mL - 1 ug/mL) in complete DMEM for 4h OR for 24h (although I am aware that a long priming step might induce TLR tolerance mediated by IL-10 secretion).

Cells were then washed and Pyrin inflammasome was induced by TcdB (100g/mL or 200 ng/mL) for 2h30' or for 6h. TcdB stimulation was done either in complete DMEM media or in Opti-MEM,

but any combination of the conditions above was able to lead to inflammasome activation, assessed by secreted IL-1b levels. 

I will be grateful if anyone can help me set up this experiment.
