I worked with P. aeruginosa PAO1 for quite a number of years already. First, in my lab at the California State Polytechnic University, I used an ATCC strain. Now I use a strain that Dr. Barbara Iglewski sent to a colleague.
In both cases, the strain produced a lot of mucus. Have you found mucoid variants of this strain? I know that hydrogen peroxide can induce a mucoid conversion of the strain but, do they convert spontaneoulsy to mucoid variants? I can attach pics if you would like to see them.

Already established that pseudomonas nearly always mutates into a mucoid strain. The alginate-containing matrix of the mucoid strain is thought to allow the formation of protected microcolonies and provide increased resistance to opsonization, phagocytosis, and destruction by antibiotics. Its mutation which increase the adaptation capability of strain. It can be virulent pulmonary pathogen causing cystic fibrosis, be careful !

Karen, thanks for your response. It is well known that it mutates into a mucoid strain in cystic fibrosis patients and in the presence of hydrogen peroxide. But I haven´t seen any paper describing a spontaneous mutation in vitro. In Pseudomonas biofilms alginate does not contribute to biofilm formation and is less abundant that the other two exopolysaccharides indeed. I haven´t seen any paper on spontaneous mutation in vitro. If you know one, please let me know. Thanks.
Ok I will try to find some information about spontaneus mutation. I can say from my experiance that during long term crioconservation of stain with the use of liquid nitrogen some of pseudomonas strains became mucoid. After that I make and suspension of starin with very smal concentration 10^10or 12 In phisiological solution and keep in thermostate. After that I use membran filtratuon technique for isolation. Some of starins became normal not mucoid.
Does the conversion between mucoid and non-mucoid phenotype require a genetic mutation?
Could anybody tell me, What is the ATCC number of Pseudomonas aeruginosa 01 (PA01) strain?
