Hello everyone.
I am a student and I have done a research about the regeneration of protoplasts from carnation. I made a protocol with sugar, ms and 2,4D-. I only used an auxin and the protoplasts divided. When I used an auxin and a cytokinin they did not divide.
I search on the internet how this can be, because plants need those hormones. But I can't get an answer so I thought maybe some of you will know.

It is possible that the auxin that you used in your protocol, 2,4-D, is promoting cell division in the protoplasts by activating the cell cycle, and that the cytokinin is not necessary for this process. Additionally, the cytokinin may be inhibiting cell division by counteracting the effect of the auxin.
It is also possible that the cytokinin you used is not compatible with the protocol or the type of cells you are working with, and that it is causing some type of inhibitory effect on the protoplast regeneration process.
It is worth mentioning that the hormonal balance of a plant cell is dynamic and complex, different hormones interact with each other and with the environment to regulate different processes, so the results of your experiment should be interpreted with caution.
