I was wondering if anyone has a detailed protocol of how to determine the number of bacterial cells in a culture sample. A protocol that includes how to do the calibration/growth curve and how to use this to determine the number of cells in the sample (with calculations if possible) .
I will be doing a invasion assay with staphylococcus aureus and need to know the number of bacterial cells in my sample to see if I need to dilute to get a MOI of 10. An OD600 = 1 is recommended but not sure how this is correlated to bacterial cell number

Hi,
OD600 does not directly relate to bacterial cell number, but rather to the biomass. Therefore you would want to generate a standard curve to correlate cell count to OD600.
