Hi every body
I want to oligomerize my monomer protein that is 75kDa, with using 0.5% glutaraldehyde in final concentration. I tried to detect its dimer form in 37C temperature for 3 min. and visualized it on 6% SDS-PAGE, but I couldnot see any bands in this condition in lane 3,5,7 according to below picture.
Can you help me about it?

If the protein is highly cross-linked, it will not enter the gel. It may be seen as staining at the edge of the wells and/or in the stacking gel. I think I see evidence of this in your gel picture in lanes 5 and 7, counting from the left.
You may want to try reducing the glutaraldehyde concentration.
