I do reductive alkylation at up to 15 mg/ml without problems using dimethylamine borane and formaldehyde.

It depends on the behavior of your protein anyway.

I am working on Immunoglobulin, DTT or 2-ME was used as reducing reagent and Iodoacetamide was for alkylation. I am not going use formaldehyde due to keep other protein's bioactivity. Thanks.
