Do Qualitative analysis of the samples first and then do quantitative analysis. This will help you to determine what is the phytochemical content of the extract. then you are supposed to take up purification of the components of the extract using chromatographic techniques and then do all the tests to say that its purify and determine the physical and chemical properties.

First of all you need to know the family of the plant under your study, try and find out some compounds previously isolated from other species of the same family. Knowing that will help you get some tips of the classes of phytocompounds that might be present in your plant. The experiment you performed will not be enough to elucidate compounds, instead you need to run complete ID (1H and 13C) and 2DNMR (COSY, HMBC, HSQC, NOESY etc) together with UV, IR, GCMS/HPLC which will help you elucidate the right compounds. Remember, knowledge of organic spectroscopy is crucial.

Hi, You have done LC-Qtof MS, Did you used HRMS? If yes, than you can have exact MW of your compounds (by comparing also with the adducts, the MW can be confirmed), than predict the molecule formula using isotope pattern; you have exact MW, Molecular formula, MS and MS MS now you can use the data to predict your compounds  by Metfrag, Metlin, Massbank or other on-line data bases; for confirmation you need standard(s), or you can do NMR (1,2D) of your compound(s).

Your LC-qTOF data should allow for identifying compounds that have already been described for the plant species, in case that meaningful fragmentation data and/or UV-spectra are present to support matching exact monoisotopic masses. The databases Gunawan mentioned are helpful. Be critical about data base matches, e.g. flavones can hardly be distinguished from isobaric anthocyans based on MS and MS/MS data. Unequivocal identification requires NMR experiments with pure compounds. However, NMR data of the crude extract might be helpful with regard to main constituents.

3. Extract the aqueous fraction with petroleum ether (nonpolar compounds)

4. dichloromethane (nonpolar compounds)

5. Ethyl acetate (Compounds with intermediate polarity)

6. Butanol. (Polar compounds)

Remove the solvent from each extract and analyze the fractions. For nonpolar compounds use GC-MS.

you need to separate the components of the mixture before to do analisys like NMR, FT-IR or microelemental analisys. Sequential extraction will serve, but consider also to do column chromatography in normal phase (silica) or size exclusion chromatography (sephadex LH-20 or toyopearl). If you can, make semi-preparative or preparative HPLC. After HPLC indicated that the sample is pure then performs the analysis as 2D-NMR

Dereplication is a great idea. My understanding is that dereplication allows you to throw out known compounds or structure classes so that you are not wasting time re-inventing the wheel. Perform dereplication as early as possible in the process. The question is how- literature search and a relevant bioassay should help.

you should separate the compounds by column chromatography. at first use the TLC and then column chromatography, after that HPLC and for determination of compounds you should use NMR.

I think you should proceed with preparative HPLC both polar and nonpolar column. Collect each fraction. Do the bio-assay invitro first then invio. Identify the mass spectra by LCMS for polar and GCMS profiling for Non-polar volatile compounds. Then you will get preliminary idea about  phytochemicals. Do esterification for non-volatile. Then do the analysis of desired sample like  FTIR, H NMR, C NMR. You will elucidate possible structure with bio-activity.

the best way of phytochemical analysis for a crude extract of plant material is to go for column separation, in order to get fractions and then follow NMR.

Preliminary phytochemical screening using standard methods for qualitative and quantitative determination of compounds such as flavonoids, saponins, tannins, phlobatanins, phenols, cardiac glycosides etc may not be out of place. HPLC and GC-MS also help to identify the actual compounds present, with their percentage. If you are carrying out solvent-solvent partitioning, you could use different solvents in their order of increasing polarity to partition e.g. ethanolic extract can be partitioned with n-hexane, chloroform/dichloromethane and methanol, butanol etc

do preliminary screening for qualitative analysis for saponins alkaloids terpenoids and flavonoids.. use methanol mostly for quick search of the TLC profiles

General screening of all phytochemicals would be necessary.
