I am working on developing a method for phytate analysis (IP6) using the mass spectrometry detector by tune method to detect the parent ion and precursor ions. The following concentrations of 50, 100, and 200 µg/ml of the IP6 standard (Purity 98%) were directly injected into the mass spectrometry, and the peak of interest was detected at 659 m/z in the negative ionization mode, however, using the fragmentation (ms/ms), the parent ion and precursor ions were not detected at 0, 20, 25, 30 collision energy. There was no signal. I do not know what could be the reason for this. Any recommendations for what I can do? Please see the photos below.
