What is the specifity of the FC-regents for determination of tanninic content after extraction? Up to me it is a non specific method measuring the total phenolic concentration. Does this include both tannins and lignin or even worse, just lignin alone?
The Folin-Ciocalteu method is not without drawbacks because it is not specific only to polyphenols. The Folin-Ciocalteu reagent is extremely sensitive to the reduction of hydroxyl groups of other molecules such as reducing sugars, vitamin C and some proteins etc., possibly present in the sample to be analyzed, which makes it possible to obtain results overestimated. Finally, the determination of phenolic compounds is "a very difficult problem that has not yet received definitive solutions". This remark of Ribereau-Gayon (1968) still retains all its meaning despite the many works done in recent years.
