I am doing lipofectamine transfection. Still no working. I have a doubt regarding the purity of DNA. My pDNA was obtained from Midiprep, but in the last step I diluted too much. To fix that I did DNA precipitation.

The protocol for DNA  precipitation is simple:

I took 2ml of DNA (from my midiprep with low concentration because was too much diluted) into a microtube

  • Add 200 uL of 3.0 M pH 5.2 sodium acetate
  • Add 1.4 ml Isopropanol room temperature, mix well.
  • Centrifuge 10,000-15,000 g for 30 min.at 4C.
  • Resuspend the pellet with  100 uL- 150 uL ddH2O.
  • Messure DNA concentration.
  • However, I am concern if doing this could affect my future experiments, like transfection with lipofectamne, because salt contamination or something related.

    My pDNA had the following results from the spectrophotometer:

    A 260 =28.522

    A280=15.2

    A260/280=1.67
