I am having trouble with my Oil Red O stain for my rat MSCs. What is the best aqueous mounting medium to use? I followed the protocol from IHCWorld (http://www.ihcworld.com/_protocols/special_stains/oil_red_o_ellis.htm) and the Oil Red O solution was bought from Sigma. Under light microscopy it looks like there are lipids vacuoles but they are not staining red. Any idea why?
The protocol you followed is correct. May be, you can try "OIl O red solution" from different company like EM sciences (26504-01Oil Red O Solution). Also use a control tissue which has lipids. Are you looking lipids staining after adipogenesis of MSCs or simply MSCs alone. I doubt if simply MSCs alone will give you visible red staining. That is my opinion. Let us hear from others.
I’m using cell culture cells not fresh tissue. What’s the protocol for them?
I used a glycerol in 1X PBS in ratio 9 glycerol : 1 1X PBS.
Normal medium that use in routine hematoxylin and eosin is not work for oil red o.
