I would like to know the potential of HCT116 cells for developing subcutaneous tumor in nude mice. I want to perform a study in which I am plannng to inject 5 X 10^6 HCT116 cells subcutaneously on the dorsal side of 10 nude mice. Can anyone tell how many mice may response to tumor development (means out of 10 mice how many may develop tumor generally)??
We've done HCT-116 xenografts on a commercial basis (http://altogenlabs.com/xenograft-models/colon-cancer-xenograft/hct116-xenograft-model/) and they do tend to grow in a stable manner with good growth curves as well. One million cells in matrigel is the right dosage, and if you do everything correctly you should get tumor growth in all mice. Assuming that you're having a relatively stable experimental setup and you're using immunodeficient mice, there should be no reason for tumors to not form.

HCT116 cells should form tumors in nude mice with no general problem (usually).
Make sure viable cells are dispensed in a volume of 100uL of media and a suitable matrix (e.g Matrigel).
Depending on your endpoint 5million cells may be on the larger side of cell counts. I recommend a pilot with fewer mice to study tumor growth dynamics to find out how rapidly they grow. This is needed because nude mice will start developing skin lesions as tumor volume tends to grow ~1000mm^3. This is easily reached within about 20 days.

Hi. Rehan I will suggest to take about 6 mice and more carefully inject cell suspension. Hope visible tumor will be grown in all the mice between 14 to 20 days.
