I know LCR ligase Chain reaction (Abbott) but it's an old method.I very much prefer PCR which usually includes all the controls (positive, negative and amplification and extraction control -Roche, Qiagen etc... Concerning Chlamydia you have to decide if to detect plasmidic or chromosomal DNA or both. Additionally you can find available multiplex PCR tests in the market which contemporary detect Both the organisms and more (Mycoplasmata). Which population and samples you have to screen?? research or epidemiological or clinical studies??
thanks for the kind reply. We have vaginal swabs stored in -80. So as far as i know we cant culture them. I need to see the presence of NG/CT. I have protocols for both chromosomal DNA. But here I have just one swab, so need to follow the same procedure for both the extractions. Which one do you think is effective?Chromosomal or plasmidic DNA? And its for research purpose. It would be great if you could provide any one protocol for both the extractions , ofcourse after I could find a way to get the positive control. Roche procedure is way costlier for me!
in which country do you live? For the DNA extraction i suggest to buy Qiagen Tissue kit, which will extract all the DNA Ct plasmidic and chromosomal and Gonococcus DNA. All in house PCR are not standardized and not good for epidemiological research in my opinion.
