You have two different issues in your objective. The first is the disruption of the endometrial tissue, which can be difficult itself. The second is disrupting the Mycobacteria. This will require either bead beating or sonication as detergents won't work alone. If you have a bead beater, I would suggest using small beads (100 micron) for the bacteria mixed with larger beads that will help disrupt the tissue. The tissue contains significant levels of collagen and will flocculate,
Thanks Dr. Burden for your reply. I am totally agree with your view that its difficult to disrupt the endometrial tissue biopsy. But QIAgen DNA mini kit were used alone in this regards for so many years without any discrepancy. I just wanted to know if anybody has used that kit that whether to proceed the same as per the protocol mentioned for tissue sample given on page no. 32-33 of QIAgen dna mini kit or to treat with the instruction mentioned on page no. 56 given for dna extraction from gram positive bacteria with preparing lysozyme lytic solution first.
