We are working on mycolactone and we would like to ionize mycolactone on a MALDI-TOF/TOF mass spectrometer.
We have already tested several common matrices such as HCCA, SA, sDHB, 9AA, DAN and a mixture of HCCA and DAN.
However none of these matrices are able to ionize our mycolactone. The difficulty is that the m/z of mycolactone is below 800, so it near or in the matrix range.
Is someone has a protocole to share?

Matrix range is not an issue as this molecule has very different isotopic pattern from matrix, which should be easy to distinguish after collecting signal in reflectron mode and modeling isotopic clusters. Besides, you can always subtract spectrum of matrix blank. Poor ionization is an issue. Perhaps, increased concentration or use a different MS technique will help. Sometimes I need to go to ridiculous 200 -300pmol of material on spot in order to obtain signal from poorly ionized compounds. Good luck!
