I am facing problem regarding purification of anabaena. i have used serial dilution and plate streaking method. But everytime the culture is contaminated with chlorella vulgaris. I have to perform DNA extraction. Please guide. Thanks in advance
spread ur diluted sample on agar plate(2/3), observe them for isolated anabaena colony...or take diluted sample n pick up anabaena cells under inverted microscope with pasteur pipette
If the specific strain you are working with forms long filaments, you could try performing serial dilutions of your cultures and then selectively filtering your samples with membranes that allow solely Chlorella to pass through. Finally, the membranes should be thoroughly rinsed with culture medium and cultivated again.
In case the filaments are short or too sensitive to the shearing induced by filtration, I would suggest you to develop a very dilute culture and pick out the Chorella cells using a micro pipette and an inverted microscope, starting a new culture from that drop you have placed on the slide.
