Dear all,

I am trying to immortalize mouse microglia using lentivirus produced in HEK293T cells (pCAG-VSVg+pCMV-dR8.2 dvpr+pHAGE-SV40). However, transduction is not efficient, and all cells die after selection. Transduction work on other cells (mouse myofibroblasts), so I know everything is okay with the virus. However, microglia have many mechanisms to save themselves from infections and foreign DNA. Does anyone have any advice on improving transduction on microglia (I think the same goes for any other immune cells)?
