In a 2 by 2 Mixed Factorial Anova, how would you go about conducting post hoc tests when you only have two groups. Would you conduct an independent samples t-test after the Anova to explore significant interaction effects?

A 2x2 mixed factorial model is such that one factor is fixed effect and the other factor is random effects. If the main effect of the fixed effect factor is significant then a post-hoc test may be done to compare the levels. If however the main effect of the random effect factor is significant no post-hoc test is necessary. The interactions are tested in the ANOVA directly.
