I have to do postpartum milk DNA extraction for my project using QIAGEN kit. I use Qubit flurometer and also the Nano to measure the DNA concentration. Matter of thing is, don't know why the outcome is poor as follows ;
0.304(Qubit flurometer) ,
2.3 (NanoDrop),
0.046(A260),
0.007 (A280),
6.50 (260/280),
0.12 (260/230)
I will appreciate your suggestion to improve my result.

I don't know why its so necessary to extract genomic DNA from milk? as you know, milk is a platform for proteomic studies since a wide variety of proteins are embedded within its matrix. However, you can use blood as an excellent alternative. I suggest to extract gDNA from blood manually using my recent universal method for DNA extraction. I'm sure you will extract DNA sufficient to perform at least 1000 PCR experiments. Kindly download this protocol by this url:

Our research participants are post partum lactating women and we are trying to use their milk sample for the DNA extraction that is required for the subsequent experiments. I am trying to avoid all kinds of contamination steps while extracting. But still I am not satisfied with the outcome and looking for a better solution.
Concentration of DNA in milk is bound to be low. Dont believe on absorbance guided quantitation. You should ideally check the presence of DNA directly by performing PCR.
