How to determine the drug dosage or concentration and number of days to administrate the drug to induce immunosuppresed rat. The study will focus whether the extract boost or supprese the immune system. Please advise and thank you in advance.

There is extensive reporting regarding the use of cyclophosphamide (CY) in animal models. The dose and regimen used will impact the type and level of suppression induced. My experience in mice for inducing a severe suppression is to use three i.p. doses (day -1 infection and days +1 and +3 post infection) at 180 mg/kg of body weight (J Immunol, 1996, 156: 246-255). In part, we measured suppression by monitoring neutropenia and counts of other leukocytes in the peripheral blood. Others have used a single treatment of CY at a higher dose such as 300 mg/kg. You should be able to find rat-specific protocols in the literature or use the dosing similar to mice as a starting point since it is based on body mass. I hope this helps.
