Most of the published studies used proteases inhibitor during blood collection to prevent breakdown of oxytocin. I am also wondering since I did not collect the blood with aprotinin would I still be able to measure oxytocin? I intend to measure the effect of one of the psychosocial trial that I am involved in, thanks in advance!
Hi Kheng, blood is loaded with proteinase inhibitors. About 15% of all plasma proteins are proteinase inhibitors.
I don't think adding aprotinin to plasma will do much of a difference.

Oxytocin is a nonapeptide and consequently a very labile molecule. That is why good practices regarding its assay always recommend to use EDTA tubes containing aprotinin to take blood. Alternatively/also, it is recommended to keep your blood tubes at low temperature and even pre-chilled them before the blood drawing. You perhaps performed that.... Additionnally, the most important thing regarding oxytocin assay is definetely to perform an extraction (generally via SPE on a C-18 column) of plasma oxytocin before your assay (RIA or EIA): see the article enclosed for more details.
