I am triying to understand better the receptor tolerance and sensitization in dopamine D3 receptors for diferent dopaminergic agonists. Drug tolerance is defined as a loss of responsiveness to an agonist after continued exposure. We are founding some tolerance behavior but sometimes the effect after tolerance develope is lower than control. May it be explained as an "hipertolerance" that affects basal responsiveness? ¿Any clue about what may happen here?

Dear Alejandro, if I understood correctly, you are getting a lower response to dopaminergic agonists in cells previously exposed to those drugs when you compare it to non-exposed cells. Is this the case? If so, what can you tell about the number of receptors in both cases? Have you checked if there was, for example, a down-regulation (reduction in total number of receptors)? I don't know what cells you are using, but if this is the case, it may take some time to replace D3 receptors after continuous exposure to those agonists. Regards
Receptor downregulation is a possible explanation for what was being observed.
For D3R, I guess there Pharmacological Sequestration Occur. which kind of agonist at which concentration you used that is also important. if you can describe your question in more details, then might be easier to answer.
