I want to know the encapsulation of my drug and for the blank (autozero) I measured the absorbance of the solution or emulsion of poloxamer and gelatin without drug, but the absorbance insn't stable, this changed when I did autozero... and I don't know if it's the gelatin that causes interference with the UV spectrum.
At what wavelength u r taking absorbance? if u r taking below 254 nm , it may not be stable , because excipients causes interference
the solutions is transluscent, I'm taking the absorbance at 227 nm (that it's the wavenumber of the drug)
Aqueous solution is transluscent, so at certain wavelength you could see your dissolved substrate inside
Gelatin, i.e. its amino acids have high absorption in the UV. Especially working at 227 nm is very sensitive to interferences.
