Generally, silencing or transient knockdown is easier through use of RNAi. But I’m afraid the stable survival of cell line is really dependent on the function of the gene you are trying to over-express or knockdown, or whether there are other redundant genes that can compsensate the loss or balance the effect some of its function that is essential for viability.
