We use enzymatic digestion.

The aorta is cannulated, and via a Langendorff coil, retrograde perfused with various solutions. Importantly, one solution contains a collagenase and a protease. This essentially digests the ECM, allowing the myocytes to be separated by gentle agitation.

This protocol (usually) provides cells suitable for patch-clamping.
