I would appreciate if anyone can suggest me the general isolation methods for isolating beneficial bacteria from soil and the best soil which has promising beneficial bacteria as well as the different media for culturing the bacterial isolates.

Most soil bacteria are non pathogenic and the most easiest and simple method to culture soil bacteria is the plate culture method where a simple media like Nutrient agar can be used to isolate total heterotrophic bacteria. While if you have specific requirement to a particular bacteria you use any selective media to culture like in my on instance I isolate hydrocan degrading bacteria using oil agar

The guidance offered by Dr. Singh and others is excellent for getting your studies underway. I would confirm that both types of target microorganisms and sources for initial isolation are keys for consideration in developing your experimental approach. My best examples are from work in seeking soil- and rhizosphere-based bacteria for potential weed biocontrol. We looked at a range of sites under contrasting management that was thought to offer a similar range in microbial types and activities. Sharifah , your question is very pertinent , in the context , what shall be possible source of promising microbes..? It will be most prudent to use , preferably the native isolates and crop -based. when you visit the field , you find certain portion of filed has an excellent crops stand , dont you feel this is the portion of the field , where from soil samples are to be taken , instead of portion of the field having poorer crop stand . Likewise if you are inside an orchard , lets look at the those trees having excellent fruit yield with quality with any infestation pests or diseases , identify those trees for sampling and isolation of most promising microorganisms.
You may collect the soil samples either from the crop rhizosphere (root surrounding areas or from the best part of the field as Dr. Srivastava has suggested ) and go for dilution plate technique using the following media like Ashbys Mannitol agar for Azotobacter isolation, Stephens medium for Nitrosomonas and Nitrobacter sp., Yeast -extract mannitol agar for Rhizobium isolation, Barrs medium for sulphur bacteria etc.

I would just like to add some more selective isolation media of beneficial microbes from rhizosphere soil apart from what Tarafdar Sir has mentioned. They are: Okon's B medium for Azospirillum, Pikovskaya's agar for Phosphate solubilizers and Pseudomonas Isolation agar for Pseudomonas isolates.

My question is addressed to Dr Tarafdar and other colleagues: We conventionally follow the serial plate dilution method by pour plate where visual counting of each and every is a big problem , we can use colony counter , image analysis etc ...is there some other methods available for precise colony counting .
It is also claimed, serial plate dilution method counts only the culturable microbes..?
What are the latest developments in studying the soil microbial load ..measuring optical density is one of them....
Yes, you can measure through optical density as well as microbial biomass.

The question you put forward is interesting. Regarding latest development's, I would like to make mention that recently I have heard of an app for iPhone and iPad which can count cfu load in a short time. This app is called CFU SCOPE. But, I don't really know it's potentiality over our traditional system of serial dilution. Information about the app is available in Google.
