I have been performing platelet isolation from PRP and prepare the platelet extracts using lysate buffer. Now, I want to isolate the platelets from PRP and store the the whole platelets for future electron microscopy. I would much appreciate if you suggest the appropriate steps for this procedure.

We work with platelets in our lab also, and when we have prepped platelets for EM in the past, we immediately embed fresh platelet pellets (versus freezing them at -80, etc). I suggest this particularly if you're looking at keeping platelet membranes intact. This would entail, depending on the protocol you're using, treating with glutaraldehyde, osmium tetroxide, propylene oxide, etc., and you'd ultimately have an embedded pellet you could slice, stain, and view with the EM.
