For my experimental observations, I need to know the greenness index (from green leaf to yellow) of the leaf of the plant Vigna unguiculata. This plant also has a pest known as Aphis craccivora which is brown to black in colour. I am looking for software, that can be used to get both spectrums based on the digital images of these organisms.

I'm thinking this can be done using OpenCV (in Python) and possibly looking at the histogram of colour frequencies in the image. There are a lot of assumptions here however, about quality-of-light, angle, how badly the pest has infected the plant, etc

You can use MATLAB and their image analysis toolbox. The software requires a licence though and a bit of experience in coding. Further more, to make sure you capture the correct colours in relation to the lighting conditions you need a ColorChecker / MacBeth chart while taking the photographs.
