I am looking for a protocol to assay Phosphorylase, Phosphofructokinase and Hexokinase activity in insects. Because of high toxicity of 2-Mercaptoethanol, I don't want to use this material.
2-Mercaptoethanol is used to protect enzyme-thiol of an eventual oxidation. It is possible to assay enzymes without this compound, as the oxidation process (if any) is not as rapid as it is supposed. Besides, dithiothreitol or even cysteine can be used instead of mercaptoethanol.
