Ion selective electrode (Cu2+) is useful for determining free Cu+2 ions in extracts of unbuffered salts (i.e. CaCl2).used for enviromental analyses in soils. By the way, thanks Robert for the paper.
ICP-OES or ICP-MS are laos options if you have access to the instrumentation
You can determine Cu activity in soil solution, which is the most direct measure of Cu toxicity for plants and microorganisms. See attached pdf.
ICP-OES and ICP-MS are excellent options, but the price of instruments (and their maintenance) can be quite expensive for some people/departments.
Voltammetry (electro-analytical technique) is relatively inexpensive and it can also be used for samples/matrices with relatively higher content of salts.
