I am involved in studying the hippocampus of rats. To study the histology of the hippocampus region of the brain, is there a specific formula for sectioning the tissue? For example, how many 50-micron-thick sections and how many 5-micron-thick sections should be made to get the exact area of the hippocampus? My goal is to examine the exact same area of the hippocampus for all groups and slides.

the easiest way to find out which is the rgion of interest, is to cut the whole brain and collect every section. If you are using fixed frozen sctions (50µm thickness )I would recommand to put all sections in microtiter 96- well plate. Take one section of each row, mount it on a slide and stain it with Cresyl violett or HE. You can do this even when you take cryo- sections (5µm) directly on the slide. Here I recommand to take after every 3rd or 4th slide one section on an extra slide for CV or HE staining. Precise dicribtion of the slides is essential otherwise you won't find your sections any more. After staining you can find the correct area of your region of interest.
