The THP-1 cell showed the classical monocyte (CD14++ CD16-) behavior. If we want to study the non-classical CD14-Cd16++ without drawing blood and isolate the monocyte with FACS, what can we do?
the cell line suggested sounds very appealing. However, it should be noted that in addition to the few early papers by Dialynas et al no futher publications are available on this cell line. So likely this was not a stable line. Of note, in the discussion the authors state that "2MAC presently requires irradiated PBL for passage." It is very well possible that the monocyte properties noted in the paper stem from the irradiated blood monocytes that have been added to the cultures regularly.
Unfortunately there is no human cell line available at this point, which represents non-classical monocytes. Attempts to mature the most advance cell line (Mono Mac 6) to become CD16-positive have not been successful thus far.
So for the time being you will have to work with primary blood monocytes.
