i have to measure protein concentration of bacteria on polyethylene surface. for this i used coomassie brilliant blue solution in perchloric acid as an indicator. but acetic anhydride is not available in our lab. could someone suggest me the alternative of acetic anhydride.
You can make it from dry Sodium Perchlorate and concentrated HCl via an exchange reaction: NaClO4 + HCl (conc, in excess) = HClO4 + NaCl (precipitate). Afterwards, you can filter out NaCl precipitate and remove the excessive HCl by gently heating the filtrate in a fume hood or on open air. HCl is much more volatile than HClO4, and can be removed nearly quantitatively with little loss of HClO4. The resulting HClO4 solution contains only traces of NaCl and HCl, and is suitable for the majority of biochemical applications, which require HClO4. The detailed protocol is attached.
