Do you use a more convenient and more straightforward method to separate
leukocytes in flow cytometric determination of anti-leukocyte antibodies?
In our practice, we use phycol-verographine separation solution (ρ
=1,088g/cm3).
What we see as a problem is that the method requires four blood samples: 3 for
direct immunofluorescence (for separation) and 1 native for indirect IF,
which is difficult to obtain, especially from young children.

Dear Peter,
What kind of leukocyte do you use? (Neutrophil or lymphocyte). You need patient blood just direct IF. You can use healthy donor cells and patient serum for indirect one.
