I have a very complicated particle surface including amphiphilic molecules with PEG. I expect to have hydroxyl groups on my surface when my particles are in water. What I try to do is, I try to change the hydroxyl groups with carboxylic acid groups using carbonyldiimidazole. Then, I use these molecules on my particle surface exposing COOH groups so that I can use them for further reaction. However, I don't have a good way of determining if and how many COOH groups I have on the surface. Is there an assay that can help me figure that out?
