Hi. I am dealing with stacked tiff data for optical mapping of heart.
to make sure good ratiometric calculation, I think I need reliable background subtraction onto each images.
At first, I subtracted the intensively low-passed images onto each frame, but it gave me a not good one.
So can you suggest the good approachs for ratiometric (I'm using Di-4-anepps for voltage sensitive ratiometric imaging) imaging? or at least just reliable background-substraction scripts for matlab?

I suggest to try the background correction MATLAB script from Vincent Mazet:

It was written to remove background from 1D signals, not images.
You can use it independently on each row / column of your image.
Or you can compute the 1D average of your image: mean(image,1) and then compute the average background.
Or you can extend the script to 2D.

You may use the Top-hat filtering from matlab (imtophat, https://www.mathworks.com/help/images/ref/imtophat.html). With using the following code, the function is expected similar with 'rolling-ball background substraction' (I compared the Matlab results with that of ImageJ using rolling-ball background subtraction):
se = strel('disk',5); % Create the disk structuring element
im=imtophat(im,se); % Subtract background
