I am a master’s student and currently working on trying to find the best parameters to conduct an alpha-glucosidase inhibition assay on plant seed extracts. I am having troubles in choosing what the concentration of the enzyme solution or the substrate (p-nitrophenyl-a-D-glucopyranoside) solution should be. Is there a detailed procedure of the alpha-glucosidase inhibition assay that I can follow or any research paper that can guide me on how to choose these parameters? I would really appreciate it if anyone could provide some information. Thank you.
