Most of the researchers are challenging against cryo-shock. So, they are generally using glycerol, which unfourtunately has a contraceptive effect on sperm. This affect decreases sperm fertility, which does not reach to demanded in vivo and in vitro results. Thus, could we prepare glycerol free extender ? Do you have any experinece ?

For mammals, the most used penetrant cryoprotector is Glycerol, but you also have avaiable many others such as amines (dimethylformamide for exemple), Ethylene glycol and others. If these Cryoprotector is still a problem for your species, you can try use techniques such as refrigeration, where sample is preserve just for a few days but there is no need of penetrant cryoprotector. You can also try to stabilish dry mantenace.

Thank you for your answer, surely some techniques are providing opportunities instead of our question@. However, we would like to cryopreserve and long-term storage at -196 C without glycerol, which is even best option.
Do you have any idea for tris formulation (including egg yolk without glycerol) ?

Glycerol is working as a cryoprotectant agent as well. You can minimize its conc through pilot experimentations...
