I wish to make PBS buffer to resemble tumorigenic pH and I have seen papers using 5 or 5.4 pH for that but I couldn't find any protocols for the same. Please help.
Phosphate has a pKa of 6.8, and hence is unsuitable for buffering at pH 5.0 (more than 1 pH-unit away from pKa). You need to use another buffering substance, suitable are for example carboxylic acids (~4.8) or pyridine (4.9).
Once you have decided on the buffering substance and its concentration (which depends on the amount of acid or base produced in your assay), you should add ions to make your buffer similar to the natural environment of your protein (high Na + Ca, low K and Mg for extracellular, other way round for intracellular proteins). In addition, sucrose, manitol and similar substances are used to adjust the osmotic pressure.
