Actually i want to go for differential expression analysis in response to a certain treatment in my selected species of wheat which has already its transcriptome available. Do i need to do sequencing of transcriptome again ? Your valuable suggestions are welcome....Thank you.

I agree with Mathew's advices but the GEO database from NCBI can offer you to get access to those data at the only condition that your request fits their data (https://www.ncbi.nlm.nih.gov/geoprofiles/).

Well, I assume the data you have come from 2 different conditions otherwise as Mathew pointed, you can make any DGE. The data you have has to match exactly the conditions we want to experiment, the sequence has to be of high quality (for explicit study). The sequencing platform is as well important depending on the focus of your research. I guess a study can not be an iteration of an old one however you can use the published data to 'confirm/refer/duplicate' your new study.
